Can't be done, at least in the way you describe. Brush definitions don't look at color - only intensity on a grayscale level. They can't look at color when being defined, because they have to work with whatever color you pick when actually using the brush.
You can try a dual brush setup. Define your custom brush as the black spiral. Then select a white hard round brush of your choice. Select dual brush in the brushes panel (F5), and select the spiral brush. I don't really know what effect you're going for, but you can experiment with different blending modes and other settings in dual brush to see if you can get what you want.
If the above doesn't work, I want to know what exactly you're trying to do. I may be able to tell you an entirely different approach to do it.
